Reducing Small RNA Ligation Bias – Small RNA Library Prep Solutions for Illumina® Sequencing

Small RNA sequencing is ideal for quantitative profiling of small RNAs (microRNAs, piRNAs and snoRNAs).  It is used to analyze tissue specific miRNA expression patterns, disease associations and to discover previously uncharacterized miRNAs. However, ligation specific biases reduce the ability of small RNA-seq to accurately quantify microRNA. These biases are mitigated through the incorporation of adapters with degenerate bases during library preparation.

PerkinElmer’s line of NEXTFLEX® Small RNA Kits for Illumina® sequencing are ideal for the conversion of these small RNA transcripts into barcoded cDNA libraries for next-generation sequencing on Illumina® sequencing platforms. With the incorporation of randomized adapters to reduce bias and AIR ligase to increase ligation efficiency; the NEXTFLEX® Small RNA kit v3  offers better data quality and a greater proportion of productive of sequence reads than competitive small RNA library prep protocols.
